建立一株人源性骨肉瘤细胞系并进行生物学测定 ,为骨肉瘤研究提供良好的实验模型。无菌切取骨肉瘤手术标本 ,于裸鼠表面包埋接种成功后移植瘤标本体外培养得到细胞株 ,应用形态学观察、流式细胞仪、生长曲线测定、肿瘤标志物测定等方法进行生物学特性研究。结果建立了一株具有肺转移特性的人源性骨肉瘤细胞系PL 1,其成瘤率为 10 0 % ,潜伏期为 12天 ,肺转移率为 10 0 % ;取其 32代细胞进行生物学特性测定 ,发现其形态结构、排列方式、分化程度与原发瘤保持一致 ,染色体形态为人类核型 ,众数在 6 4~ 6 6之间 ,流式细胞仪检测G1期细胞为 5 2 1% ,碱性磷酸酶及骨形成蛋白抗体免疫组化染色为阳性。说明应用裸鼠体内移植建立的人骨肉瘤细胞系PL 1与原发瘤保持相同的生物学特性 ,为一个稳定的细胞系。
The establishment of a human osteosarcoma cell line and biological determination of osteosarcoma provide a good experimental model. The specimens of osteosarcoma were excised aseptically and transplanted in nude mice on the surface of the tumor. The transplanted tumor specimens were cultured in vitro to obtain the cell lines. The morphological characteristics, flow cytometry, growth curve and tumor marker determination were used to investigate the biological characteristics the study. Results A human osteosarcoma cell line PL 1 with lung metastasis was established. Its tumorigenic rate was 100%, the incubation period was 12 days and the lung metastasis rate was 100%. The 32-generation cells were used for biology It was found that the morphological structure, arrangement and differentiation of tumor cells were consistent with those of primary tumors. The chromosome morphology was human karyotype with a diversity of 6 4 ~ 6 6. Flow cytometry showed that G1 phase cells were 5 2 1 %, Alkaline phosphatase and bone morphogenetic protein antibody immunohistochemical staining was positive. It shows that PL 1, a human osteosarcoma cell line established by in vivo transplantation in nude mice, maintains the same biological characteristics as the primary tumor and is a stable cell line.
