目的 :探讨 p2 7kip1 、PCNA及 p1 6与胃肠道类癌转移的关系。 方法 :采用免疫组化SABC法检测 2 5例胃肠道类癌组织中p2 7kip1 和PCNA蛋白表达 ,同时用PCR法检测 p1 6基因的缺失情况。 结果 :p2 7kip1 在胃肠道类癌阳性表达率为 64 % (1 6/ 2 5) ,PCNAⅠ~Ⅱ级和Ⅲ~Ⅳ级表达率分别为 56 %、44 %。 2种蛋白阳性表达率与分化程度无相关性 (P >0 0 5) ,与淋巴结转移相关 (P <0 0 5)。p2 7kip1 与PCNA表达负相关。p1 6基因纯合性缺失率为 48% (1 2 / 2 5) ,其缺失率与类癌分化程度和淋巴结转移无关。结论 :p2 7kip1 低表达、PCNA高表达与胃肠道类癌转移有关 ,胃肠道 ;类癌的发生与p1 6基因缺失有关 ,但可能为早期分子事件。
Objective: To investigate the relationship between p27kip1, PCNA, p16 and gastrointestinal carcinoid. Methods: Immunohistochemical SABC method was used to detect the expression of p27kip1 and PCNA in 25 cases of gastrointestinal carcinoid tissues. The deletion of p16 gene was detected by PCR. Results: The positive expression rate of p27kip1 in gastrointestinal carcinoid was 64% (16/25). The expression rates of PCNA Ⅰ ~ Ⅱ and Ⅲ ~ Ⅳ were 56% and 44% respectively. There was no correlation between the positive expression rate of the two proteins and the degree of differentiation (P> 0.05), and lymph node metastasis (P <0.05). p2 7kip1 was negatively correlated with PCNA expression. The homozygous deletion rate of p1 6 gene was 48% (1 2/2 5). The deletion rate of p1 6 gene was not related to the differentiation degree and lymph node metastasis. CONCLUSION: The low expression of p2 7kip1 and the high expression of PCNA are associated with the metastasis of gastrointestinal carcinoid. Gastrointestinal tract and carcinoid carcinogenesis is related to the deletion of p1 6 gene, but may be an early molecular event.
