目的合成拟除虫菊酯类农药人工抗原并对其进行鉴定。方法采用功夫菊酸和3-间苯氧甲酸2种拟除虫菊酯类农药中间体,与四碳“间隔臂”——γ-氨基丁酸反应合成2种半抗原,经质谱鉴定合成成功。以牛血清蛋白为载体,采用活化酯法制备拟除虫菊酯的2种免疫原;以卵清蛋白为载体,采用混合酸酐法制备了3种包被原;经紫外光谱鉴定,粗略计算了偶联物最终浓度及结合比。结果制备的免疫原免疫Balb/c小鼠后得到的小鼠多抗血清经ELISA检测效价达1∶50 000以上,IC50达200 ng/ml。结论拟除虫菊酯人工抗原的制备成功。
Objective To synthesize and identify artificial antigens of pyrethroid pesticides. Methods Two kinds of pyrethroid insecticide intermediates of Kung Fu chrysanthemic acid and 3-isophthalic acid were used to synthesize two kinds of hapten with the four-carbon spacer “γ-aminobutyric acid” . Using bovine serum albumin as carrier, two kinds of immunogens of pyrethroids were prepared by activated ester method. Three kinds of coated protoplasts were prepared by mixed anhydride method using ovalbumin as carrier. The UV spectra were used to identify the coupling Final concentration and binding ratio. Results The immunized Balb / c mice were immunized with multi-antiserum and the titer was over 1: 50000 by ELISA and the IC50 was 200 ng / ml. Conclusion The pyrethroid artificial antigen was successfully prepared.
