目的:建立高效液相色谱法测定鬼箭羽药材中4种主要有效成分儿茶素、去氢双儿茶素A、香橙素和柚皮素的含量。方法:采用DiamonsilTMC18(250mm×4.6mm,5μm)色谱柱,以甲醇-乙腈-0.1%甲酸水溶液为流动相梯度洗脱(0~35min,9∶9∶82→17.5∶17.5∶65),流速1mL·min-1,检测波长280nm(儿茶素、香橙素和柚皮素)、380nm(去氢双儿茶素A),柱温30℃。结果:儿茶素、去氢双儿茶素A、香橙素和柚皮素线性范围分别为0.428~3.424μg(r=0.9998),0.255~1.53μg(r=0.9992),0.0788~0.630μg(r=0.9996),0.026~0.208μg(r=0.9996);平均加样回收率(n=6)分别为100.5%(RSD=3.5%),101.5%(RSD=2.3%),97.33%(RSD=1.1%),97.81%(RSD=2.7%)。结论:9批样品测定结果表明本法简便、准确,重复性好,可用于鬼箭羽药材中儿茶素等4种成分的含量测定。
OBJECTIVE: To establish a HPLC method for the determination of catechin, dehydro-catechin A, orange-tannin and naringenin in Ghost-Arrow feather medicinal materials. METHODS: Diamonsil TMC18 (250 mm × 4.6 mm, 5 μm) was used to elute the mobile phase of methanol-acetonitrile-0.1% formic acid (0-35 min, 9:9:82 → 17.5:17.5:65) Min-1, detection wavelength 280nm (catechin, orange and naringenin), 380nm (dehydrogenated catechin A), the column temperature 30 ℃. RESULTS: The linear range of catechin, DHC, orange and naringenin were 0.428 ~ 3.424μg (r = 0.9998), 0.255 ~ 1.53μg (r = 0.9992) and 0.0788 ~ 0.630μg (n = 6) were 100.5% (RSD = 3.5%), 101.5% (RSD = 2.3%) and 97.33% (RSD = 0.9996) 1.1%), 97.81% (RSD = 2.7%). Conclusion: The results of nine batches of samples show that the method is simple, accurate and reproducible. It can be used to determine the content of four components of catechin in ghost-feather herb.
