目的 :探讨具有抗氧化作用的纯阳子对兔髂动脉球囊成形术后血管局部 c- myc基因表达的影响及c- myc表达的高峰期与血管壁解剖结构之间的关系。方法 :32只日本大耳白兔分成 4组 (每组 8只 ) :A组 :左侧髂动脉为假手术组 ,右侧髂动脉为正常对照组 ;B组 :双侧髂动脉球囊成形术组 ;C组 :球囊成形术加维生素 C和 E组 ;D组 :球囊成形术加纯阳子治疗组 ,均于术前用药 7d。应用原位杂交的方法 ,观察纯阳子对球囊损伤后 2 h血管局部 c- m yc基因表达的影响及其与血管壁解剖结构的关系。结果 :A组左、右髂动脉均未见 c- m yc基因的表达。B组近管腔部可见一层阳性显色 ,中膜沿平滑肌细胞排列方向可见阳性显色。C组及 D组均见近血管腔部有一层阳性显色 ;中膜沿平滑肌细胞排列方向也见一些散在的表达 ,两组之间无明显差异 ;但与 B组相比 ,颜色明显变浅。结论 :纯阳子能抑制兔髂动脉球囊损伤后 c- m yc基因的过度表达 ;球囊损伤后 2 h(损伤后 c- myc基因表达的高峰期 ) ,c- myc基因在靠近管腔处表达最显著。
Objective : To investigate the effect of pure neutron with antioxidation on local c-myc gene expression after balloon angioplasty in rabbits and the relationship between the peak of c-myc expression and the anatomical structure of vessel wall. Methods: 32 Japanese white rabbits were divided into 4 groups (8 in each group): Group A: left iliac artery was sham-operated group, right iliac artery was normal control group; Group B: bilateral iliac artery balloon-shaped Surgery group; group C: balloon angioplasty plus vitamin C and E group; group D: balloon angioplasty plus pure sun treatment group, both preoperative medication for 7 days. The in situ hybridization method was used to observe the effect of Chunyang on the local c-myc gene expression after 2 h of balloon injury and its relationship with the anatomical structure of the vessel wall. Results: There was no expression of c-myc gene in the left and right radial arteries of group A. A positive coloration was observed in the proximal luminal region of Group B, and a positive staining was observed along the arrangement of smooth muscle cells in the media. In both groups C and D, there was a positive staining near the lumen of the blood vessels; some scattered expression was observed along the direction of the smooth muscle cells in the media, there was no significant difference between the two groups, but the color was significantly decreased compared with the B group. . CONCLUSIONS: Chunyangzi can inhibit the overexpression of c-myc gene after balloon injury in rabbits; 2 h after balloon injury (peak of c-myc gene expression after injury), c-myc gene is close to the lumen. The most significant expression is at the office.
