目的优化穿心莲种子无菌萌发的实验条件。方法以植物组织培养技术,采用单因素法考察采收时间、种子的成熟状态、光照条件、预处理、消毒时间、培养基对穿心莲种子萌发的影响。结果穿心莲种子萌发的优化条件为:贮藏时间半年、完全成熟的种子、有光照条件、40%硫酸浸泡2 h预处理、消毒5 min,培养基为MS或1/2 MS。结论采用该优化条件可以在短期内培育大量整齐一致的穿心莲试管苗,且不受季节和环境条件的限制,具有较强的实用性和推广性。
Objective To optimize the experimental conditions of the seed germination of Andrographis paniculata. Methods Plant tissue culture techniques were used to investigate the effects of harvest time, seed maturity, light conditions, pretreatment, disinfection time and medium on seed germination of Andrographis paniculata. Results The optimal conditions for the seed germination of Andrographis paniculata were as follows: half-full-time, fully-matured seeds were pretreated with 40% sulfuric acid for 2 h and disinfected for 5 min. The medium was MS or ½ MS. CONCLUSION: This optimized condition can be used to cultivate a large number of uniform and in vitro testicular plantlets of Andrographis paniculata in the short term without being limited by the seasonal and environmental conditions. Therefore, it has strong practicability and promotion.
