伊马替尼是一种2-苯胺嘧啶衍生物,除对 bcr/abl 融合基因阳性的慢性粒细胞白血病(CML)和急性淋巴细胞白血病(ALL)细胞有明显的作用外,还可以抑制 c-kit 酪氨酸激酶。在急性髓系白血病(AML)中 c-kit 被异常磷酸化。我们用不同浓度的伊马替尼对 c-kit~+AML 患者骨髓细胞进行体外处理,采用 MTT 法,RT-PCR 法及流式细胞术探讨其对骨髓细胞凋亡的影响,旨在为临床应用伊马替尼治疗 AML 提供理论依据。
Imatinib is a 2-aniline pyrimidine derivative that has a significant effect on cells with chronic myeloid leukemia (CML) and acute lymphoblastic leukemia (ALL) positive for bcr / abl fusion gene but also inhibits c- kit tyrosine kinase. C-kit is abnormally phosphorylated in acute myeloid leukemia (AML). We used different concentrations of imatinib on c-kit ~ + AML patients with bone marrow cells in vitro treatment, MTT assay, RT-PCR and flow cytometry to explore the impact on bone marrow cell apoptosis, aimed at clinical Imatinib treatment of AML provides a theoretical basis.
