目的评价尼古丁对L-929细胞毒性及作用机制。方法采用(MTT)法检测尼古丁对L-929细胞增殖的影响,采用流式细胞术观察尼古丁对L-929细胞周期的影响。结果不同浓度尼古丁作用于L-929细胞24,48和72 h后,均能抑制L-929细胞生长。0.01,0.1μg/mL尼古丁作用24,48 h细胞毒性为1级,作用72 h细胞毒性则为2级。10 000μg/mL尼古丁作用24 h细胞毒性为3级,作用48,72 h细胞毒性则为4级。随着尼古丁浓度增加,G0G1期细胞比例逐渐增高,而S期和G2期细胞比例则呈下降趋势。结论尼古丁对体外培养L-929细胞具有明显细胞毒性,且阻遏细胞周期。
Objective To evaluate the cytotoxicity and mechanism of nicotine on L-929 cells. Methods The effect of nicotine on the proliferation of L-929 cells was detected by MTT method. The effect of nicotine on the cell cycle of L-929 cells was observed by flow cytometry. Results Nicotine could inhibit the growth of L-929 cells at 24, 48 and 72 h after L-929 cells were treated with different concentrations of nicotine. Cytotoxicity of 0.01,0.1μg / mL nicotine for 24,48 h was grade 1, and cytotoxicity was grade 2 for 72 h. Cytotoxicity of 10 000 μg / mL nicotine for 24 h was grade 3, and cytotoxicity was grade 4 for 48 and 72 h. With the increase of nicotine concentration, the proportion of G0G1 phase cells gradually increased while the proportion of S phase and G2 phase cells decreased. Conclusion Nicotine has obvious cytotoxicity on L-929 cells in vitro and suppresses the cell cycle.
