Objective:1. To explore if ECL (Enhanced ChemiLuminescene)direct nucleic acid labelling and detection systems can be used in the fingerprinting of pig genomic DNA;2. To detect the inbreeding degree of banna miniature pigs with DNA fingerprinting and STR.3. To study the genetic background of the three strains of minipigs with DNA fingerprinting and STR.Methods:Clearly and highly polymorphic DNA fingerprint maps of Banna,guangxibama,guizhou miniature pigs were obtained by southern hybridization with a nonisotopically HRP(Horseradish peroxidase) labelled multiloci minisatellite probe JL-02.and 3’HVR,detected by enhanced chemiluminescene with endonuclease HInf I or HaeIII,In addition,to detect the genetic background with STR(short tandem repeat).Results:1. We acquired the following available DNA transfer and hybridization conditions.In brief, 10 μg genomic DNA was digested with 40u Hinf I or Hae III endonuclease in 30 μ 1 total reaction volume for 4h at 37 ℃ .After digestion,the clearly DNA fingerprint was achieved.2. inbreeding degree was obtained with probe JL-02. The fragment numbers above 2kb with probe JL-02 ranged from 13-17, the similarity coefficient of various individuals from family "805" - "807" in substrain JB and family "111", "121", "133", "151" in substrain JS was 0.92 ± 0.04, 0.85 ±0.13, 0.86 ±0.02. 0.80 ±0.05, 0.84 ±0.09, 0.88 ± 0.08,which is higher than that between families(0.46 ~ 0.65).Furtherly,similarity coefficients of family "805" in substrain JB and family "151" in substrain JS were 0.92 and 0.88 respectively.3. inbreeding degree was obtained with probe 3’HVR. Corresponding, The fragment numbers above 2kb with probe 3’HVR ranged from 6-15, the similarity coefficient of various individuals from family "805", "807" in substrain JB and family "111", "121", "133". "151" in substrain JS was 0.94±0.09. 0.85±0.09, 0.84 ±1.7. 0.78 ± 1.6, 0.85±0.42. 0.87 ± 0.07,which is higher than that between families(0.46 - 0.65).Furtherly,similarity coefficients of family "805" in substrain JB and family "151" in substrain JS were 0.92 and 0.88 respectively.4. endonuclease Hinf I or Hae III digested DNA fingerprint was obtained for guangxibama miniature pigs. The fragment numbers above 2kb endonucleased by HinfI and Haelll for GuangXiBama miniature pigs are 11.7 and 24.5 respectively, the similarity coefficient of various individuals of Guang Xi Bama miniature pigs with endonuclease Hinf I and HaeIII are 0.54 ± 0.07 and 0.55 ±0.08 respectively;5. endonuclease Hinf I or Haelll digested DNA fingerprint was obtained for guizhou miniature pigs. The fragment numbers above 2kb endonucleased by HinfI and HaeIII for Gui Zhou miniature pigs are 15.8 and 17.6 respectively, the similarity coefficient of various individuals of gui zhou miniature pigs with endonuclease Hinf I and Haelll are 0.53 ± 0.04 and 0.53 ± 0.07 respectively;6.we Acquired Allele constructure,AIlele Frequencies, Allele Heterozygosity for three laboratory miniture pigs.Conclusions:1. ECL can be used to DNA fingerprinting of pigs and the same effect can be achieved to those generated from the isotopically labelled probe.2. The two probes are available to the DNA fingerprinting for Banna miniature pig and obtain the same result.3. The similarity coefficient in families is higher than that between families.4. similarity coefficients of family "805" in substrain JB and family "151" in substrain JS were the highest.this showed that the two families have the higher inbreeding degree, smaller genetic difference among individuals and the better homogeneity.5. various endonuclease will produce various DNA fingerprint fragments,but generate the same similarity coefficient.This also showed that the two kinds of miniature pigs reach the demand of closed colony.6.STR is suitable to Genetic Monitoring for three laboratory miniture pigs.
