Epigenetic regulation of gene transcription by RNA-directed DNA Methylation in rice

  DNA methylation acts as a stable epigenetic modification and plays many important roles in higher eukaryotes.In dicot Arabidopsis,the establishment of DNA methylation at CG,CHG and CHH sites (H=A,C or T) is through the RNA-directed DNA methylation (RdDM) pathway.However,the molecular and biological functions of conserved RdDM factors in monocots remain elusive.Here,we systematically characterized the functions of RdDM homolog factors in rice,including Ospol4a,Osrdr2,Osdcl3a,Osago4a,Osago4b and Ospol5.Our data show that DNA methylation on the transposable elements (TEs),especially MITEs (Miniature Inverted repeat TEs),is actively controlled by RdDM.Importantly,knock-down mutants of these factors caused dwarfism,and even sterility,indicating that DNA methylation plays essential roles during rice growth.Consistent with this,genes significantly up-regulated in mutants were enriched in several biological processes,such as cell wall macromolecule catabolic process and cell wall organization.Our data suggest that RdDM extensively contributes to the transcriptional regulation of gene expression during rice development.
