Studies on Chinese cabbage diseases focused on downy mildew,soft rot and virus disease,however a new Verticillium wilt disease became more and more serious for the past years in Beijing,Yunnan and many other district.The infected plants displayed yellow-leaf,brown vascular,stunted growth and eventually plant death,which cause a servere decrease in agricultural production and qualification.In this study,4 fungi isolates from 4 different infected plants were obtained,the pathogens proved to be V.dahliae by morphological characteristics,pathogenicity test and rDNA-ITS sequence analysis.The high virulent strain BCHW 10-2 was used for pathogen host range test.Studies revealed that BCHW 10-2 caused severe Verticillium wilt diseases of many crops,including turnip,radish,cabbage,chinese kale,broccoli,cauliflower,savoy,tomato,pepper and eggplant.Further,we designed primer pair HW1-F/HW1-R to amplify the pathogen,which matched specially rDNA-ITS sequence of V.dahliae.The real-time PCR assay enabled sensitive quantification of V.dahliae in 4-week-old seedlings with a sensitivity limit of 5×10-3 ng·μL-1.Most of all,the detection results of Q-PCR perfectly consisted with phenotypic identification.Thus a real-time PCR as a reliable molecular diagnostic approach was established to assess V.dahliae infection level and identify materials for verticillium wilt resistant.It will be useful for genetic improvement of resistance to verticillium wilt with a more sensitive and less time-consuming method for breeders.
