Sedative and hypnotic effect of new N6-(4-hydroxybenzyl) adenine riboside modifier YZG-330

  OBJECTIVE To investigate the sedative and hypnotic effect of YZG-330.METHODS Open-field apparatus were used to determine the sedative effect of YZG-330.Loss of the righting reflex was used to determine the start of sleep.Sleep latency and sleeping time were evaluated in this experiment.Then we observed its effects on sleep-wake profiles in mice by electroencephalogram (EEG) analysis.Results: YZG-330 decreased the locomotor activity in a dose dependent manner.At the dose of 0.125, 0.5, 2 mg·kg-1 i.g., compared with vehicle, YZG-330 decreased the total locomotor activity by 11% , 48 % (P<0.01), 69% (P0.05), 68 % (P < 0.001).YZG-330 (0.125,0.5, 2 mg·kg-1 i.g.) increased the sleeping time by 25%, 64% (P < 0.01), 500%(P <0.001), respectively.Each dose of YZG-330 did not affect sleep latency significantly.Intracerebroventricular (i.c.v.) infusion of YZG-330 could induce sleep in mice directly.The result of EEG analysis shows that YZG-330 administered at doses of 1 or 4 mg·kg-1 (i.p.) significantly increased the amount of non-rapid eye movement (NREM) sleep (77.9% , P < 0.05 ; 134.2% , P < 0.001, respectively) and decreased the amount of WAKE and rapid eye movement (REM) sleep in a period of 4h.Conclusion: These results indicate that YZG-330 has sedative and hypnotic effects, and suggest its potential application of insomnia.
