针-药结合对肝星状细胞中PDGF-βR/ERK信号通路及细胞外基质的堆积的抑制作用

目的:探讨姜黄素联合针刺应用对四氯化碳导致大鼠肝纤维化模型PDGF信号通路蛋白和mRNA表达的影响,为临床治疗肝纤维化提供基础.方法:采用CCl4造模法制作大鼠肝纤维化模型,通过使用姜黄素给药的方法联合针刺太冲、期门、肝枢,以及电针足三里,进行联合治疗,同时设空白对照组、模型组、针刺组和姜黄素组作为对照分别进行干预.造模结束后分离肝组织,采用ELISA法检测血清中PDGF含量;western blotting检测HSCs中PDGF-βR、ERK、JNK、P38、α-SMA、collagen、Fibronectin、MMP-9和TIMP-1蛋白的表达;real-time PCR方法检测HSCs中PDGF-βR mRNA的表达。结果:ELISA检测结果显示,与模型组相比,针刺组、姜黄组、针药结合组均能有效地降低大鼠血清PDGF的含量,且针药结合组作用最为显著;与模型组相比较,姜黄素联合针刺组能显著下调PDGF-βR,ERK,α-SMA,collagen,Fibronectin,MMP-9蛋白的表达以及PDGF-βRmRNA的表达。结论:姜黄素联合针刺通过抑制CCl;导致肝纤维化大鼠PDGF通路,减少胶原沉积,促进ECM降解,起到治疗肝纤维化的作用。
