iTRAQ-based quantitative proteomic analysis reveals the serum biomarkers in acute ischemia stroke

  Objects:The purpose of this study is to identify serum biomarkers which can assist the neuroimaging evaluations to diagnose acute ischemia stroke.Methods:Six cases of AIS with hypertension were selected and matched with six healthy controls.lsobaric tags for relative and absolute quantitation(iTRAQ)coupled with MS used to compare and analyse differential protein of these 12 samples respectively.Enzyme linked immunosorbent assay(ELISA)validation was undertaken in 25 independent samples.Results:In total,242 proteins were identified from 49297 spectras using iTRAQ coupled with MS.Compared with the healthy control group,there were 14 differentially expressed serum proteins,13 0f them were up-regulated,and one of them was down-regulated.Furthermore,CST3 and PCYOXl were used to carry out the validation of western blot,and the results were consistent with the result of iTRAQ coupled with MS.Conclusion:KIC9,APOH,PCYOXl,IGJ,PIGR may be novel and potential biomarkers for early diagnosis of AIS.

目的:为观察脑电双频指数(BIS)应用于猕猴麻醉深度监测的可行性与有效性.方法:4-6岁猕猴8只,雌雄各半,体重5-7kg,术前夜禁食水.将猕猴进行绑定后,前额部剃发并用酒精擦拭,将BIS电极片的3号电极贴于眉弓外2cm,其余3个电极以1号电极尽可能位于前额正中进行固定.固定好的猕猴同时监测血压、心率、血氧饱和度监测等生命体征.开放静脉后,采用异丙酚输注行静脉麻醉(生理盐水1∶1稀释),初始输注速
目的:脓毒症是临床常见的危重症,急性肺损伤(ALI)是其严重并发症,死亡率较高.线粒体功能障碍与脓毒症关系密切.线粒体自噬可选择性降解损伤的线粒体,维持自身稳态.近年来发现氢气对多种疾病具有治疗作用.本课题组前期研究表明氢气可减轻脓毒症小鼠ALI,但其具体作用机制及靶点尚不十分清楚.本研究拟观察氢气吸入对脓毒症小鼠肺组织自噬相关蛋白LC3,线粒体自噬蛋白pink/parkin表达的影响,探讨其与线
