The damaging effects of hyperhomocysteinemia on the brain

  To investigate the damaging effect of hyperhomocysterinemia (HHcy) on cerebral cortex and hippocampus in apolipo-protein E-deficient (ApoE-/-) mice,we established mouse models of HHcy by feeding a high-methionine diet for 16 weeks.Plasma concentration of homocysteine was detected in each mouse.Brain structure changes were observed by optical and transmission electron microscopy.The activation of astrocytes in the cerebral cortex and hippocampus was evaluated using glial fibrillary acidic protein (GFAP) immunohistochemical staining and western blot analysis.After treatment with folic acid & vitamin B12,plasma concentration of homocysteine was significantly increased (P < 0.01),the number of pyknotic neural cells (P < 0.05),and GFAP expression in astrocytes of cerebral cortex,hippocampus,and cerebrum were significantly decreased (P < 0.05) in HHcy ApoE-/-mice compared to those in C57BL/6J and ApoE-/-control mice.These results indicate that folic acid and vitamin B12 treatment reversed the inhibition of HHcy to GFAP expression in astrocytes.Taken together,HHcy caused neuronal injury and decreased GFAP expression in astrocytes in ApoE-/-mice.Treatment with folic acid and vitamin B 12 could alleviate HHcy-caused brain injury.
