AIM:To observe the expression of NF-κB p65 and MMP-2 on murine viral myocarditis model.METHODS:Six-week old inbred male mice were randomly assigned to control(n=25)and myocarditis group(n=30).The myocarditis and the control groups were inoculated intraperitoneally with 0.1ml 10-5.69TCID50/ml CVB3 or vehicle(PBS)alone respectively.Eight mice were sacrificed 4 and 10 days after injection.Blood and heart specimens were harvested for measuring the content of serum TNF-αand MMP-2,NF-κB p65 和IκBα in cardiac tissues.The concentrations of TNF-αwere detected by ELISA.Western Blotting was performed to analyze the contents of MMP-2,NF-κB p65 and IκBα in myocardium.RESULTS: In the myocarditis group,the expression of MMP-2 and NF-κB p65 protein in cardiac tissues were significantly higher than those in the control group(P<0.05);and the concentration of TNF-α in blood were higher than those in the control group(P<0.05);the expression of IκBα protein were lower than those in the control group(P<0.05).CONCLUSION:TNF-α,NF-κB p65 and MMP-2 were higher in mice with acute viral myocarditis.The increased expression of them might be involved in the pathogenesis of viral myocarditis.
