Comparative analysis of stability and hydrolysis of Aconitine by liquid chromatography-mass spectrom

  Aconitine is a typical diterpenoid alkaloid of medicinal interest, which can be found in many plants such as Aconitum carmichaeli Debx.In this article, a method was developed aiming to the comparative analysis of stability and hydrolysis of Aconitine in different solvent using liquid chromatography coupled with electrospray ionization ion-trap mass spectrometry (LC-ESI-MS).Aconitine was resolved in distilled water, ethanol and acetonitrile with different pH value including 2.0, 7.0 and 10.0, respectively.These solutions were designed to store at-20℃, 0℃, 20℃ for 7 days or heated to boil for 30 min in order to test the stability of Aconitine in different solvents under different conditions.With the experimental results, no hydrolysate was detected in all of the solvents with pH 2.0 while only one hydrolysate was found in all of the solvents with pH 10.0.Moreover, the stability of Aconitine was stronger both in ethanol and acetonitrile than in distilled water with pH 7.0, and there were more hydrolysates could be detected in distilled water stored at 20℃ or boiled than stored at low temperature.By comparing the molecular mass, retention time and full-scan MS2 spectra with those of Aconitine, four hydrolysates were detected, which were elucidated as Anhydro aconine, Aconine, Deacetyl Aconitine and Acotine according to the relative reports in references, respectively.
