Aim To study the effects of Polygalasaponin F (PGSF) on PC12 cells. Methods The viability of cells was determined by MTT assay.Cell apoptosis was assessed by propidium iodide/Hoechst 33258 staining and flow cytometry assay. The expression of Bcl-2 and Bax proteins were detected by Western blot. Results PGSF elevated cell viability markedly. In oxygen-glucose deprivation and reperfusion model (OGD/R), it improved the morphology of cells, attenuated apoptosis of cells and prevented the up-regulation of Bax as well as the down-regulation of Bcl-2.Conclusion PGSF could protect PC12 cells against injury induced by serum deprivation, oxidative stress or OGD/R.
