Effect of C-Type Natriuretic Peptide on Maturation and in vitro Fertilization Embryo Development of

  Abstract C-type natriuretic peptide (CNP) is a peptide hormone belonging to the family of natriuretic peptide and chiefly expresses in ovary.This study aimed to demonstrate the effect of C-type natriuretic peptide on maturation and in-vitro fertilization (IVF) embryo development of buffalo oocytes, basing on the expression of its cognate receptor NPR2 in buffalo follicle.The results are shown as follows:(1) Immunohistochemistry and Real-time PCR technologies were used to detect the expression of NPR2.The result showed that both NPR2 protein and mRNA were present in buffalo cumulus-oocyte complexes (COCs).(2) Oocytes were assessed after COCs pretreated in maturation culture medium containing 0, 50, 100, 200, 400 nM CNP for 6h respectively and then cultured in common medium until 24h.The result showed that adding with 200nM CNP into the maturation culture medium could significantly improve the oocyte maturity rate (P<0.05), the blastocyst rate of the subsequent IVF embryos (P<0.05), and the cell numbers in blastocyst (P<0.05).In conclusion, NPR2 mainly expresses in COCs of buffalo follicle.The suitable concentration of CNP (200nM) is good for the efficiency of buffalo oocytes in vitro maturation and the acquirement of embryos developmental potential.
