蛋白酶体调节子PTRE1参与生长素信号调控

  生长素在植物生长发育中发挥重要调节作用,其主要通过生长素受体TIR1介导负调控因子AUX/IAAs的降解,进而释放ARFs转录因子调控下游基因表达.泛素/蛋白酶体(Ubiquitin-Proteasome)途径可以有效快速地选择性的移除相关蛋白,在激素信号响应、发育及胁迫响应等过程中发挥重要作用.虽然已有研究表明了蛋白酶体在生长素信号中的重要作用,但是其调控机制以及AUX/IAA降解的平衡机制目前仍不清楚.我们鉴定了一个动物中蛋白酶体活性抑制蛋白PI31的同源基因,PTRE1 (PROTEASOME REGULATOR 1),其编码一个脯氨酸寓集蛋白.功能研究表明PTRE1介导了生长素调节的蛋白酶体活性过程并协调TIR1介导的蛋白降解以调控生长素信号.PTRE1定位于细胞膜、细胞核及内质网,生化分析表明PTRE1可以促进26S蛋白酶体活性.突变体ptre1表现出多重发育缺陷并对生长素响应不敏感;遗传研究表明TIR1与PTRE1具有协同效应,ptre1突变体中TIR1介导的AUX/IAA降解减弱,同时生长素可以通过PTRE1抑制蛋白酶体活性.进一步的研究表明生长素调控了PTRE1在细胞膜及核中的分布从而调控其活性.这些结果为了解AUX/IAA的降解调控及生长素作用机制提供了新的线索.
