Development and Application of an Immunoassay for Detection of Cd Based on Specific Nano-body Agains

  In this study,p-SCN-Bn-DTPA was chosen as a bifunctional chelator to facilitate the synthesis of antigens for Cd,and the complete antigen Cd-DTPA-BSA/OVA was gained successfully.The phage antibodies Cd-DTPA-VHH 4-18 and Cd-DTPA-VHH 6-15 were produced through phage display technology successfully and Cd-DTPA-VHH 4-18 was the best.The relative affinity is 5.95×108 L/mol,there exists no cross-reactivity between the antibody and the carrier protein BSA/OVA and also the non-metallic antigen DTPA-BSA/OVA,and almost no cross reaction with other metal ions.The initially established indirect competitive ELISA immunoassay method detection for the detection of Cd with the prepared phage antibody met the national standard of water quality monitoring.The lowest detection limit for Cd using the phage antibody VHH 4-18 was 0.2nM,and the spiked results showed that the mean recovery was more than 90%.
